    On September 14, 2015, Rainer Weiss ’55, PhD ’62, began the first day of his vacation in Maine by doing what he does every morning after breakfast, wherever he is: he checked the experiment logs for the Laser Interferometer Gravitational-Wave Observatory (LIGO). An announcement that weekly repairs planned for the next day had been canceled struck Weiss, a professor emeritus of physics, as odd. Moments later, a flurry of e-mails directed him to a website with images that left him thunderstruck: LIGO, the instrument he’d conceived of four decades earlier, had recorded a signal on one of its first test runs after a major upgrade. He let out a yell that brought his wife and son running. “Then the disbelief began,” he says. “The signal was too large and too perfect. It took several days before I truly began to believe that it was a real event.”

After months of analysis and rechecking of data, an international team of researchers led by scientists from MIT and Caltech announced in February that the signal was from two massive black holes that had collided 1.3 billion years ago. That collision had released a burst of power estimated to be as much as 100 times that of all the stars in the universe, producing a ripple in space-time recorded by LIGO. It was the first direct detection of the gravitational waves that Einstein predicted a century ago.

Where does the story of LIGO begin?
It started here in 1967. I was asked by the head of the teaching program in physics to give a course on general relativity. By the time 1967 rolled around, general relativity had been relegated to mathematics departments. It was a theory of gravity, but it was mostly mathematics, and in most people’s minds it bore no relation to physics. And that was mostly because experiments to prove it were so hard to do―all these effects that Einstein’s theory had predicted were infinitesimally small.

Einstein had looked at the numbers and dimensions that went into his equations for gravitational waves and said, essentially, “This is so tiny that it will never have any influence on anything, and nobody can measure it.” And when you think about the times and the technology in 1916, he was probably right.

The big thing that’s happened in the last 100 years is, people discovered things in astronomy which were very different from what they knew in 1916―tightly compact sources [of gravitational waves], enormously dense, like a neutron star, and black holes. And there was technology for doing precision measurements, because you had lasers, masers, electronics, computers, and a whole bunch of stuff people didn’t have in 1916.

So the technology and knowledge of the universe made it possible, by the time we got into this, to contemplate trying to look for gravitational waves.

In the 1960s, Joseph Weber at the University of Maryland had the idea that maybe the technology had gotten to the point where you could look for gravitational waves, and he invented a method for doing that. He imagined a sort of xylophone made of massive resonant bars. He expected that a gravitational wave would come along and pull on one of the bars and squeeze it, and as the wave left, it would leave a pulse, and the thing would ring and you could hear it.

It was the first idea that you should do something active to go look for gravitational waves. And he had claimed a discovery in the 1960s.

When I taught my course, the students were very interested in finding out what [Weber’s experiment] was. And I’ll be honest with you, I couldn’t for the life of me understand the thing he was doing. That was the problem. Because it completely countered every intuition I had now developed about general relativity. I couldn’t explain it to the students.

That was my quandary at the time, and that’s when the invention was made. I said, “What’s the simplest thing I can think of to show these students that you could detect the influence of a gravitational wave?”

The obvious thing to me was, let’s take freely floating masses in space and measure the time it takes light to travel between them. The presence of a gravitational wave would change that time. Using the time difference, one could measure the amplitude of the wave. Equations for this process are simple to write, and most of the students in the class could do it. Forget for a moment that this was a thought experiment requiring impossibly precise clocks. The principle was okay.

I didn’t think much more of it until about a year later, when I began to realize something about Weber’s experiments: nobody was getting the answer he was getting. He had made a huge and powerful claim. And I began to realize maybe this was wrong, and maybe even his idea of how it works was wrong.

So I sat down in a little room in Building 20, the “Plywood Palace” on Vassar Street, and worked the whole summer on the idea that I had talked with my students about. And knowing what you could do with lasers, I worked it out: could you actually detect gravitational waves this way? And I came to the conclusion that yes, you could detect gravitational waves, at a strength that was much better than what Weber was looking for.

What did it take to bring this idea into physical form?
We were building a 1.5-meter prototype in RLE [the Research Laboratory of Electronics] using [military] funding, and were fairly well along. All at once funding was gone, due to the Mansfield Amendment, which was a reaction to the Vietnam War. In the minds of the local RLE administrators, research in gravitation and cosmology was not in the military’s interest and support was given to solid-state physics, which was deemed more relevant. For the first time, I had to write proposals to other government and private agencies to continue our research.

Nobody was seriously working on gravitational-wave interferometry yet, although as I learned later, others had thought about it as well. A German group at the Max Planck Institute in Garching had just been through building a Weber bar. They had worked with the Italians and found that Weber was wrong. They had probably done the very best experiment of anybody to show this. That was the mid-’70s.

They were asked to review my proposal to the National Science Foundation, just as they were thinking about the next thing to work on. They had been thinking, as many other groups in the world had at the time, to make even better Weber bars by cooling them to close to absolute zero. Instead they made a decision to try the interferometer idea. They called me to ask if there were any students that had been trained on the 1.5-meter prototype so that they could offer them a job. (At exactly the time they called there were none; a little later David Shoemaker, who had worked on the MIT prototype, did join the Garching group.) They then built a three-meter prototype, got it working, and did a beautiful job.

Next they built a 30-meter one. Somewhat later a group in Glasgow, Scotland, directed by Ronald Drever, who also had built a Weber bar, began working on interferometric detectors.

By the time I got funding from the NSF and got going again, the German group had really solved most of the technical problems with the idea and shown that all the calculations I had done were right on the money, that it worked just as calculated. They also added some ideas of their own that made it better.

At the LIGO detector in Livingston, Louisiana, it took 40 days to extract over two million footballs’ worth of air from two four-kilometer-long vacuum chambers, resulting in one-trillionth the air pressure at sea level.
A key step was in 1975. Because I was also doing studies in cosmic background radiation supported by NASA, I was asked by NASA to run a committee on uses of space research in the field of cosmology and relativity. What came out of that committee, for me, was that I met [Caltech physicist] Kip Thorne, whom I had asked to be a witness for the committee.

I picked Kip up at the airport on a hot summer night when Washington, D.C., was filled with tourists. He did not have a hotel reservation, so we shared a room for the night. We ended up spending the whole night talking about what might be interesting experiments for Caltech to do. Kip had developed one of the best theory groups in gravitation at Caltech and was thinking of bringing an experimental gravitation group there. We laid out on a big piece of paper all the different experiments one could build a new group around. I told him about this thing we were working on. He had never heard about it, and he got very interested. What came of it was that Kip and I eventually decided Caltech and MIT would do this [project that became LIGO] together.

What were some key moments that drove the project forward?
In the later 1970s, the MIT group, now including Peter Saulson and Paul Linsay, did a study with industry to determine the feasibility of building a large, kilometer-scale gravitational-wave interferometer. The study looked at how to make large vacuum systems and considered how to determine costs for scaling up the prototypes, the possible sites where one could build five- to 10-?kilometer L-shaped structures with minimum earth-moving, and the availability of optics and light sources. We looked at the possible sources of gravitational waves and several competing interferometer concepts that had been prototyped in different labs in the world. The information was put in a report, called the Blue Book, and presented to the NSF in 1983. Scientists from Caltech and MIT together presented the ideas developed in the Blue Book, as well as the results of the prototype research.

The proposal we presented was to make a detector system sensitive enough to actually detect gravitational waves from an astrophysical source (not just a new prototype). The proposal was to build two detectors. You couldn’t do science with one; you had to have two separate detectors, equally sensitive, and long enough.

That was a real struggle later on. You wanted to maintain those ideas, and people later wanted to shave it down: Why not just build one long one? Why build it so long? All these arguments were made, but we stuck it out. We had to―otherwise we would never have survived and we wouldn’t be here today. We got an endorsement from the committee: risky research with the possibility of a profound outcome, well worth considering as a new project by the NSF.

By the mid-1980s, the NSF kept trying to figure out how to start this. Then, in 1986, an interesting thing happened that finally broke the logjam. Richard ?Garwin, who had worked with Enrico Fermi [1938 Nobel laureate in physics] and with the Department of Energy, and made all the calculations and did the actual development of the first hydrogen bomb, had become chief scientist of IBM. He had read about Weber’s experiments and decided with another IBM associate to build a little one, much smarter than what Weber had built―and he saw nothing.

The NSF was trying to sell this huge new program for gravitational waves. Garwin gets wind of it, and he thought he had slayed this dragon. He wrote a letter to the NSF saying, “If you’re going to persist with this, you’d better have a real study.”

So we ran a study at the American Academy of Arts and Sciences on Beacon Street in Cambridge. It was a one-week meeting with an excellent committee of hard-nosed scientists who reviewed the research we’d done with the prototypes, the feasibility studies to make a large system, the plans for siting the system, and the cost estimates. The recommendation the committee made was unbelievably good: the project is absolutely worth doing, don’t divide it up into one detector at a time, make it the full length, no more prototypes. It also recommended a change in the management of the project to have a single director rather than a steering group, which was the way we had managed the project up to then.

By 1989, we wrote another proposal under the direction of Rochus Vogt [a former Caltech provost], which took us almost six months to write―it was a masterpiece. The proposal was to build two sites with four-kilometer-long interferometers. The interferometers were to be staged. The first detector was based on the research, now reasonably mature, from the prototypes with a sensitivity that offered a plausible chance for detection. The second detector was based on newer, advanced concepts that had not yet been fully tested but that offered the capability of a good chance for detection. The proposal made it through the National Science Board and got accepted, and money started coming in significant amounts.

By the 1990s, the rest of the history is easier. Now under the direction of Barry Barish [a Caltech physics professor], the sites were being built and developed, vacuum systems were made, and we started running the first detectors. By 2010, we had run them and made vast improvements in their sensitivity but had seen nothing. It was a clean nothing; the detectors had run at design, and we saw no anomalies that could be interpreted as gravitational waves. Based on the fact that we [had achieved our desired] design sensitivity and had carried out the science to determine some interesting upper limits on possible sources, we received funding to build Advanced LIGO.

How momentous for you is this discovery?
As far as having fulfilled the ambitions of a lot of us who have worked on this, it is momentous. It’s the signal that all of us have wanted to see, because we knew about it, we had never had real proof of it,wholesale samsung note 7, and it’s the limit of Einstein’s equations never observed before―the dynamics of the geometry of space-time in the strong [gravitational] field and high velocity limit.

To me, it’s a closure to something which has had a very complicated history. The field equations and the whole history of general relativity have been complicated. Here suddenly we have something we can grab onto and say, “Einstein was right. What a marvelous insight and intuition he had.”

I feel an enormous sense of relief and some joy, but mostly relief. There’s a monkey that’s been sitting on my shoulder for 40 years, and he’s been nattering in my ear and saying, “Ehhh, how do you know this is really going to work? You’ve gotten a whole bunch of people involved. Suppose it never works right?” And suddenly, he’s jumped off. It’s a huge relief.

Republican criticism of federal government efforts to fund new energy technologies has spilled over to ARPA-E, the U.S. Department of Energy??s popular program for backing high-risk innovations in energy. Paul Ryan, now the Republican nominee for vice president, voted last year to slash ARPA-E??s budget, and presidential candidate Mitt Romney??s energy plan says the program should shift its focus.

Such views could rule out giving money to companies like Envia Systems, a 35-person startup located in Newark, California. Following a $4 million grant from ARPA-E, it says it??s within sight of commercializing a high-capacity energy storage? technology that would cut prices for electric-car batteries in half.

??Venture capital funding took us half the way there, and the ARPA-E funding took us all the way there,?? CEO Atul Kapadia says.

The presidential campaign has revived the long-running policy debate over what role government should play in funding the development of new technologies. While almost everyone believes that it has a role in supporting basic research, the consensus breaks down at later and more expensive stages of development, such as demonstration projects. Historically, some Republicans have picked fights to keep agencies from giving grants for early-stage product research, a line that ARPA-E crossed intentionally when it was created in 2007.

ARPA-E has funded around 200 projects, all of them meant to be ??transformative?? ways to either help replace foreign oil or reduce emissions. The notion is that such projects are too speculative and risky to gain large investments from companies. ??I think it??s hard to argue the types of investments that ARPA-E is making would be made by the private sector if ARPA-E did not exist,?? says Greg Nemet, an assistant professor of public affairs and environmental studies at the University of Wisconsin.

The agency, which had a modest budget of $180 million in 2011, has many fans in Congress, including among Republicans. That means it could avoid cuts and may see its budget increase. However, last year some House members said the agency should be defunded because its projects are too commercial and sometimes replicate work already paid for by the private sector.

Critics claim one problem is that ARPA-E isn??t able to find enough research that is truly transformative. Eric Toone, the agency??s principal deputy director, calls the debate over spending a ??worthy?? one, but says most people agree ARPA-E is funding technologies at stages where there is a ??legitimate role for public investment.??

??Are we going to run out of great ideas??? he asks. ??If we keep collecting the best and brightest people of America, there??s lots to do here and there??s lots of good ideas.??

ARPA-E grants are meant to help move research ideas to the prototype or demonstration stage. Projects are given specific performance goals?Dsuch as increasing how much energy can be stored in a battery?Dthat, if achieved, would take technology a few steps beyond the best commercial products. It has financed projects such as making liquid ??electrofuels?? directly from micro?rganisms fed electricity, chemicals, and carbon dioxide, as well as a flying wind turbine (see ??Flying Windmills??) and new materials to capture carbon from coal plants.

At the agency a team of scientists actively manage research programs. It??s not uncommon for them to pull the plug if technical milestones aren??t met. Such failures are partly by design. Grants are kept small (on average, about $3 or $4 million each)?Dpart of an approach designed to pull a few winners out of a large pool of attempts.

While ARPA-E has generated a share of exciting projects, it does have one clear flaw: a lack of end customers. ??The big problem that makes energy different from most other startup enterprises is that even if you have something that works great, you probably never amass enough money to commercialize it,?? says Donald Paul, executive director of the University of Southern California??s Energy Institute and former chief technology officer at Chevron.

That??s where the Obama administration ran into trouble. The DOE tried to help some technologies toward large-scale commercialization, but after the bankruptcy of solar-panel maker Solyndra (recipient of a $535 million DOE loan guarantee), Republicans jumped in, accusing Obama of playing politics with technology. It??s become a campaign talking point: Ryan??s website calls for ??getting Washington out of the business of picking winners and losers in the economy,?? including the energy sector. Although Romney has praised ARPA-E, he??s echoed the Republican concerns by saying the agency should step back and concentrate on ??basic research.??

Such a shift would be at odds with ARPA-E??s current grant portfolio. More than a third of the agency??s grants have gone to companies (the rest go to universities and government labs), and nearly all of them are for applied research projects.

In the case of Envia, the company used its ARPA-E grant to finish development of an anode design for its prototype commercial battery pack. That wasn??t basic research: there was a clear commercial goal. ??It shortened our development time by two years,?? says Kapadia.

Envia Systems may never get its novel battery technology into vehicles. But it stands a better chance because of its ARPA-E funding. After seeing Envia??s prototype batteries, General Motors invested $7 million in the startup. During a meeting with employees last month, the automaker??s CEO, Dan Akerson, said the battery technology could be a ??game changer?? for GM, judging that it stood a ??better than 50-50 chance?? of leading to an electric car capable of going 200 miles on a charge.

He then gave what might be the perfect endorsement of ARPA-E. ??These little companies come out of nowhere,?? said Akerson. ??And they surprise you.??

Still, that Tesla was valued higher than GM underscores the profound change occurring in the global automotive industry as Silicon Valley pursues a vision for transportation -- including self-driving cars and vehicles on demand -- that could upend century-old competitors. Last week''s disappointing monthly sales results by traditional auto makers served as a further example to investors concerned that the profitable U.S. new-car market is plateauing.

"We''ve built a track record of strong financial performance," a GM spokeswoman said in an email. "We''ll stay focused on delivering outstanding results and making decisions to deploy capital where it will generate the strongest returns, to enhance shareholder value."

Tesla declined to comment.

GM is the largest auto maker in the U.S. by market share, making up 17.3% of the sales last year, according to Autodata Corp. Tesla had a 0.2% share, which beat Ferrari and Maserati.

"What''s fun about following this company now is that anything can happen, " Chaim Siegel, an analyst for Investing.com, said in an email about Tesla that aligns with investor sentiment even as the auto maker remains unprofitable and deeply in debt. "The potential is huge. The hopes are huge."

Even some of Tesla''s most optimistic followers didn''t expect the extent of the recent surge in value. "We''re pretty surprised by the recent run in Tesla''s share price to over $300 so quickly," Adam Jonas, an analyst for Morgan Stanley, wrote in a note to investors as Tesla''s market cap neared GM''s. He had been targeting a $305 price for Tesla. "Such is the power of technical factors over fundamental drivers," he said.

Tesla shares are up more than 40% this year, a move that last week led Chief Executive Elon Musk''s company to surpass Ford as the second-largest auto maker. The exuberance comes even as Mr. Musk faces huge challenges in accomplishing all he is claiming to do, including making 500,000 vehicles next year after building just 84,000 last year and creating software that would enable a vehicle to drive itself.

"It''s indicative of the market wanting to pay for potential, including into markets that don''t exist yet in any large size such as [electric vehicles], home energy generation and storage, rather than profits and cash flow today that the large auto makers generate," said David Whiston, an analyst for Morningstar Research.

Mr. Musk, who has struggled to bring out new products before, faces the daunting challenge of later this year rolling out the $35,000 Model 3 sedan, the linchpin in his plans to take the company into the mainstream from the rarefied air of selling luxury vehicles.

His acquisition of SolarCity Corp. late last year and removal of the word "Motors" from the company''s official name are part of a broader vision of being able to offer solar panels to generate energy and batteries to store that power at home or the office -- all for the benefit of the vehicles being sold. He has also begun shipping vehicles equipped with the hardware he says is needed to make them fully self-driving once the software is completed, aiming to demonstrate the prowess by year-end.

It is a vision that received a strong endorsement late last month with the revelation that Chinese technology company Tencent Holdings Ltd., best known for China''s largest social network, WeChat, had acquired a 5% stake in Tesla.

"The sooner investors view Tesla as a transportation/infrastructure company rather than just a car company, the more we believe the industry events to come over the next 12 to 18 months will make sense," Mr. Jonas wrote.

Investors continued to push Tesla shares higher last week after the announcement of a record quarter of vehicle sales. Meanwhile, GM reported a modest U.S. sales gain for March, lifted by truck demand, and said Chief Executive Mary Barra earned $22.6 million in 2016, a decline of 21% from 2015 when she was awarded a sizable retention package. Ms. Barra, at the helm since 2014, has said her goal is to make GM the most valued auto maker in the world.

For GM, Tuesday was particularly painful. The Detroit auto maker, which sold 10 million vehicles last year globally, has been rushing to develop its own self-driving technology, acquiring Cruise Automation last year in a deal that could be worth $1 billion, and investing in ride-hailing service Lyft.

Ms. Barra''s predecessor, Dan Akerson, a former telecommunications executive, was concerned about Tesla''s potential, setting up a team to study how the electric-car maker could threaten GM and challenging his senior executives to war-game-like scenarios.

GM has been especially aggressive in recent years in trying to thwart Tesla, pushing legislation in states in an attempt to block the company''s strategy of selling directly to customers instead of using franchised dealers like the rest of the auto industry has done for generations. "It''s absolutely mind-boggling that we''re even discussing GM and Tesla reaching a parallel," said Dave Sullivan, an industry analyst with AutoPacific. "It''s not as if they are sitting on some sort of blockbuster drug that isn''t available in generic form. The wide rollout of electric vehicles by Jaguar, Daimler, BMW and Audi is right around the corner."

    The real time monitoring of brain function has advanced in leaps and bounds in recent years. That’s largely thanks to various new technologies that can monitor the collective behaviour of groups of neurons, such as functional magnetic resonance imaging, magnetoencephalopathy and positron emission tomography.


This work is revolutionising our understanding of the way the brain is structured and behaves. It has also lead to a new engineering discipline of brain-machine interfaces, which allows people to control machines by thought alone.

Impressive though these techniques are, they all suffer from inherent limitations such as limited spatial resolution, a lack of portability and extreme invasiveness.

Today, Dongjin Seo and pals at the University of California Berkeley reveal an entirely new way to study and interact with the brain. Their idea is to sprinkle electronic sensors the size of dust particles into the cortex and to interrogate them remotely using ultrasound. The ultrasound also powers this so-called neural dust.

Each particle of neural dust consists of standard CMOS circuits and sensors that measure the electrical activity in neurons nearby. This is coupled to a piezoelectric material that converts ultra-high-frequency sound waves into electrical signals and vice versa.

The neural dust is interrogated by another component placed beneath the scale but powered from outside the body. This generates the ultrasound that powers the neural dust and sensors that listen out for their response, rather like an RFID system.

The system is also tetherless?the data is collected and stored outside the body for later analysis.

That gets around many of the limitations. The system is lower power, can have a high spatial resolution, and it is easily portable. It is also rugged and can potentially provides a link over long periods of time. “A major hurdle in brain-machine interfaces (BMI) is the lack of an implantable neural interface system that remains viable for a lifetime,” say Seo and co.

The difficulty is in designing and building such a system and today’s paper is a theoretical study of these challenges. First is the problem of designing and building neural dust particles on a scale of roughly 100 micrometres that can send and receive signals in the harsh, warm and noisy environment within the body.

That’s why Seo and co have chosen ultrasound to send and receive data. They calculate that the power required to use electromagnetic waves on the scale would generate a damaging amount of heat because of the amount of energy the body absorbs and the troubling signal-to-noise ratios at this scale.

By contrast, ultrasound is a much more efficient and should allow the transmission of at least 10 million times more power than electromagnetic waves at the same scale.

Next is the problem of linking the electronics to the piezoelectric system that converts ultrasound to electronic signals and vice versa. Ensuring that the system works efficiently will be tricky given that it has to be packaged in an inert polymer or insulator film (which must also expose the recording electrodes to nearby neurons).

Finally, there is the challenge of designing and building the interrogation system that generates the ultrasound to power the entire array but at a low enough power to avoid heating skull and the brain.

On top of all this is the additional challenge of implanting the neural dust particles in the cortex. Seo and co say this can probably be done by fabricating the dust particles on the tips of a fine wire array, held in place by surface tension, for example. This array would be dipped into the cortex where the dust particles become embedded.

That’s an ambitious vision that is littered with challenges beyond the state-of-the-art. However, the team has a strong background in nanoelectromechanical systems and in the interface between electronic systems and cells.

Indeed, one of the authors, Michel Maharbiz, developed the world’s first remotely controlled beetle a few years ago, a development that was named one of the top 10 emerging technologies of 2009 by Technology Review.

These guys are clearly not afraid to take on big challenges. It’ll be interesting to see how they fare.

Bucking this trend, scientists at Stanford University have synthesized an artificial DNA strand with molecules about 15 percent larger than the natural variety. This so-called xDNA has properties lacking in the puny DNA that nature cooks up. It is more stable, for example. It also glows under ultraviolet light. These traits suggest that xDNA could be useful in genetic diagnostic procedures and, potentially, artificial forms of life. “Our biggest interest is whether we can design our own genetic system,” says chemistry professor Eric Kool, who led the Stanford research team. “I think we’re well on our way.”

What makes xDNA different from regular DNA is its structure. Normally, DNA is a string of nucleotides, each of which comprises a sugar, a phosphate, and a base: either adenine, thymine, guanine, or cytosine (represented in DNA descriptions as A, T, G, and C). When DNA strands bond with one another, the bases match up in a particular way: adenine always bonds with thymine, and guanine with cytosine.

About thirty years ago, Nelson Leonard, then a University of Illinois chemist and now at the California Institute of Technology, found a way to stretch adenine so that it would fluoresce when exposed to ultraviolet light. What Leonard couldn’t do was attach the sugar and phosphate to the base, making a complete nucleotide; scientists at the time didn’t know how to make DNA, though now the process of creating artificial strands is commonly used in genetic medical diagnostics.

What Kool sought was a way of making a DNA double helix. First he synthesized two expanded bases: adenine and thymine (xA and xT). He then made nucleotides out of the xA and xT bases with appropriate sugars and phosphates. By pairing an xA with a normal T and an xT with a normal A,Cheap vaio laptops online store, Kool was able to assemble them into a double helix just wide enough to contain the stretched bases.

Building the nucleotides took four years of work. The enlarged bases increase the diameter of the helix, which needs more such steps as a result. Being larger, the molecular structure offers increased stability; while natural DNA in Kool’s lab fell apart at 21C, xDNA remained intact up to 56C. Kool’s research “shows that the double helical structure of [natural] DNA does not have to be the only one,” says Danith Ly, an assistant chemistry professor at Carnegie Mellon University and an expert in developing chemical tools for studying genomics and proteomics.

Kool’s ultimate quest for a tailor-made genetic system is a tall order-and not one to be rushed, says Ly: “In order for a biological function to exist independent of anything, it needs proteins, lipids, enzymes-all sorts of things. For us to do it in the next hundred years might be possible, but it would be difficult.” And before that, Kool must create expanded versions of the G and C bases, which will likely be comparable in difficulty?to his work on xA and xT.

Science fiction scenarios of designer genes aside, Kool believes xDNA has practical implication in diagnostics-particularly in improving the existing medical procedures that detect health conditions based on the structure of a person’s DNA. Clinicians looking for particular DNA or RNA in a person take a tissue sample and introduce an artificial DNA strand that will bond to that material. Techniques that wash away everything but bound pairs containing the artificial DNA allow labs to readily search what is left. “If it doesn’t bind properly, you know that it either a mutation or the [sought] DNA isn’t there,” says Paul Billings, vice president and national director of genetics and genomics at Laboratory Corporation of America, a diagnostics testing company in Burlington, NC.

Because xDNA bonds more strongly than regular DNA,?it would be more resilient in this testing process. Moreover, its natural fluorescence could act as a beacon, making detection easier. Even after the technique was more than a lab curiosity, however, its diagnostic utility would have to be demonstrated in the field. “First of all, you have to prove that it gets into the cells and behaves in other ways as other DNA,” Billings says. “Second, you have to prove that it’s better than other methods.” There are well-established methods that work now, and no evidence yet that the binding and fluorescent properties of xDNA would be a clear improvement.

Furthermore, the fluorescent characteristics of xDNA would likely need adjusting, says Ly. According to the paper by Kool’s team, the molecule lit up when illuminated by ultraviolet light with a wavelength of about 390 nanometers. “Tissues do not absorb very well at these wavelengths,” Ly notes, suggesting that for diagnostics use, the base would have to be adjusted to respond to light that was closer to the red end of the spectrum. But, according to Kool, this would not necessarily pose a significant problem in examinations of sufficiently thin slices of tissue. In fact, xDNA nucleotides may even change their fluorescent color or intensity when bonding to natural DNA or RNA-a phenomenon that would add more possible tools to the diagnostics kit. Certificates are supposed to verify a website as genuine to a visitor’s Web browser; that verification prevents an attacker from using a forged domain address to steal data. The certificates contain encrypted data that lets browsers and other software confirm that a website is legitimate. So by compromising the digital certificate, an attacker can pose as a secure website, such as Google’s Gmail, and intercept communications, or bypass security mechanisms and install malicious software.

“What is unusual here is not that a certificate authority was compromised, but that someone noticed,” says Moxie Marlinspike, chief technology officer and cofounder of Whisper Systems, a firm focusing on securing mobile communications. “This is happening all the time.”

The compromised certificate company, DigiNotar, is one of about 650 companies, known as certificate authorities, or CAs, that are trusted to issue the certificates. Earlier this year, another certificate authority, Comodo, acknowledged that an attacker had breached the security of its systems and issued at least nine certificates for large domains, including Google, Skype, and Yahoo. At the Black Hat Security Conference in August, Marlinspike criticized the current system of certificate authorities and offered a different model, known as Convergence, based on a peer-to-peer model of trust.

The Electronic Frontier Foundation, a digital rights group, argued in an analysis published this week that recent break-ins suggest that the choice of whether to trust a certificate authority should lay with the user, not with browser vendors or websites.

“These CAs appear to exist within around 50 countries’ jurisdictions,” the authors of the report write. “Any one of these countries could conceivably compel a CA to create fraudulent certificates for purposes of espionage or for spying on that country’s citizens.”

The latest attack demonstrates that a single breach can have far-reaching effects. A preliminary report issued by Dutch security firm Fox-IT in early September found that the intruders exploited significant weaknesses in DigiNotar’s network security, including a single account capable of controlling all its certificate servers and using a weak password for account access. The firm found that more than 300,000 unique IP addresses—almost entirely from Iran—encountered one fraudulent certificate issued for Google’s domain. Already, Apple, Google, Microsoft, and Mozilla have updated their browser to distrust any certificate signed by DigiNotar.

The Dutch government, which relies on the digital signatures issued by DigiNotar for its encrypted communications, has taken over the company’s certificate operations. In addition, it is investigating whether the focus on Iranian users could indicate that the nation’s government may have been involved in the attack.

The certificate system works, but needs increased focus on security, says Amar Doshi, a senior manager of certificate products with security firm Symantec, which acquired and now manages the certificate authority VeriSign.

“All the events of the last couple of weeks really go to show that ‘a cert is a cert is a cert’ doesn’t really apply,” Doshi says. “There are differences between certificates. There are differences between CAs.”

Some of the browser makers seem ready to focus on those differences. Last week, the Mozilla Foundation, the group that manages development of the Firefox browser, provided certificate authorities with a list of security checks to complete in eight days. It said that any authority that fails to comply with the request could find any certificates issued by them deemed untrustworthy by Mozilla.

“Participation in Mozilla’s root program is at our sole discretion, and we will take whatever steps are necessary to keep our users safe,” Kathleen Wilson, the program manager in charge of Mozilla’s CA Certificates Module, said in an e-mail to certificate authorities.?

    In October 2007, the English rock band Radiohead enhanced its already enviable avant-garde credibility by releasing its seventh album, In Rainbows, online. Fans willing to offer up their names and e-mail addresses–or at least, fake names and fake e-mail address–could pay what they chose for the album, even downloading it for free. The band, and the “tip jar” business model it had adopted, were the talk of the music press and the blogosphere for weeks.


Just days after the release, ?the site Gigwise.com, citing an unnamed source “close to the band,” claimed that 1.2 million copies of the album had already been downloaded. At about the same time, a survey by a British company called Record of the Day pegged the average price paid at about $8. But Billboard, the U.S. music industry’s leading trade magazine, estimated the number of downloads at closer to 400,000, although it accepted the $8 average. And ComScore, a consumer research company based in Reston, VA, that collects data on the online behavior of a representative two million people worldwide, calculated that in the first 29 days of October,Buy nintendo online discount store, about 1.2 million people visited the Radiohead site. Although a “significant percentage” of them downloaded the album, ComScore said, the average payment was $2.26.

As of this writing, the band has declined to release any figures. Radiohead’s manager dismissed the Gigwise report as “exaggerated.” But band representatives also called ComScore’s data “wholly inaccurate,” and Ken Kovash of Mozilla, the organization that designed the Firefox Web browser, revealed that ComScore had underestimated the number of people who visited the Mozilla site in September by about 60 percent.

Billboard surmised that a conventional CD release of In Rainbows would have brought Radiohead between two and three million dollars. If anything like the higher sales estimates obtain, then relying on the public’s largesse rather than the efficiency of record-label marketing campaigns increased the band’s take. But even if ComScore’s less impressive figures are right, and a “significant percentage” means no more than half, then Radiohead grossed $1.36 million in the last three weeks of October. That’s still a big enough number to turn some heads.

Money: It’s a Gas
The digitally compressed music file presents a conundrum that may be unprecedented in the history of commerce: soon the music industry’s chief good will be one that its customers can easily acquire at no cost.

The industry’s response to the threat of piracy has been threefold: to use digital rights management (DRM) software to limit illicit copying and distribution; to discourage file sharing through lawsuits; and to attempt to exploit the new technology in ways that preserve high profit margins.

By most accounts, the first two strategies are doomed and will eventually be abandoned. But the third has been much more successful. Last summer, iTunes sold its three billionth audio file, which means that it has generated around $2 billion in revenue for the record companies partnered with it. And while iTunes, which charges 99 cents per download, is by far the largest online retailer of compressed music files, it’s still only one of many.

As the market for music comes to be dominated by generations weaned on file-sharing software, however, piracy is likely to take a bigger bite out of online sales. So one of the questions the music industry will need to answer is how much it can afford to charge customers who consider payment entirely optional.

Data that address that question are hard to come by, but the online music store eMusic may, through a quirk of its business model, provide some guidance. The ?second-?largest online retailer of compressed audio files, eMusic still lags far behind the leader: according to its CEO, David ?Pakman, it has only a hundredth as many customers as iTunes. Those customers pay monthly subscription fees that entitle them to fixed numbers of downloads: $10 buys 30 downloads, $15 buys 50, and $20 buys 75. Subscribers who regularly download their full monthly quota thus pay from 27 to 33 cents per MP3.

But few eMusic subscribers meet that description. Since the quotas don’t roll over from one month to the next, a given subscriber’s average payment per download can fluctuate widely: 30 downloads last month may have meant an average of 33 cents per MP3, but 10 this month means an average of a dollar. “Across all of our customers and all of our usage, the effective price that consumers are paying is less than 99 cents, so we don’t believe that 99 cents is the right price point,” says Pakman. “For non-hit music, it’s probably somewhere between 60 and 75 cents a song. And that’s based on data. That’s not my gut. That’s based on what we see people willing to pay in the market.”

Pakman doesn’t believe that eMusic’s low prices are seducing anyone away from piracy, though. “Our customers are really not the piracy-prone customers,” he says. “I think that generally, piracy is the domain of youth, and we just don’t focus on the youth customer. So we don’t see piracy as eating into our ability to sell music.”

But what happens when today’s piratical youths, unintimidated by file-sharing technology and accustomed to free music, become adults?

In 1998, Bruce Schneier, a celebrated cryptographer and chief technical officer of the network security firm BT ?Counterpane, proposed an answer, a mechanism he called the street performer protocol. A band would announce the completion of a new album, and the band’s fans would begin paying arbitrary amounts of money into an escrow account. Once the total in the account crossed some threshold, the band would collect the money and release the new work into the public domain. Schneier showed that data encryption can ensure that the artist doesn’t swipe the money without delivering the goods, and that if the payment threshold is never crossed, the contributors can all get their money back (plus interest).

But Schneier himself now believes that such measures will prove unnecessary. “In real life, you’re more likely just to use trust,” he says. “Radiohead just said, ‘We’ll do it.’ They’re just relying on the good will of their fans, and their fans’ trust in them, and so on. And that just seems more plausible in our world.”

Radiohead’s reliance on the good will of its fans probably netted it more than a million dollars in the month of October, and possibly much more. But Radiohead is also the beneficiary of years of major-label publicity. In a world of unrestricted, open-?format downloads, where record companies have no way to recoup the costs of huge marketing campaigns, bands will have a harder time achieving Radiohead’s level of celebrity in the first place.

Nonetheless, smaller experiments with new methods of distribution and promotion also indicate that people are willing to pay more than they have to for music they care about. The Canadian folksinger Jane Siberry, who last year changed her name to Issa, began using the tip-jar model to sell her recordings a good two years before Radiohead did, although her website recommends that customers pay the industry standard 99 cents a song. According to statistics posted on the site, about 20 percent of downloaders pay nothing. But the average payment per song is still $1.20. Magnatune, a website that sells 569 albums by 258 artists, lets its customers choose how much to pay but sets a lower bound of $5 an album. Its 50 top albums, however, sell for an average of $8 to $10. In a slightly different vein, Team Love Records posts all of its artists’ recordings to its website as free, high-?quality MP3s. But its most popular acts still sell thousands of CDs, which is pretty good for a small independent label.

It may be that in the brave new world of Internet music distribution, rock bands will no longer generate so much revenue that they can afford to throw TVs out of hotel windows or insist that all the brown M&Ms be removed from the candy bowls in the greenroom. But Radiohead’s online release of In Rainbows contributes to the mounting evidence that musicians who build an audience will still be able to make a living doing what they love.

Aufgrund des hohen Preises und der groen Bildschirmdiagonale von 65 Zoll bietet sich als Alternative unweigerlich ein UHD-TV an. Der Hisense HE65KEC730 kostet zwar rund 200 Euro mehr, fllt dafr aber seine 65 Zoll mit vier Mal so vielen Pixeln wie das Sony-Gert. Insgesamt berzeugt er im 2D- und 3D-Bereich mit einer durchweg guten Bildqualitt. Abstriche mssen Sie allerdings bei der Tonqualitt und dem Stromverbrauch machen. Zudem fehlen dem UHD-TV derzeit Apps fr Streaming-Dienste wie Netflix oder Maxdome.

    “General and indiscriminate retention” of emails and electronic communications by governments is illegal, the EU’s highest court has ruled, in a judgment that could trigger challenges against the UK’s new Investigatory Powers Act – the so-called snooper’s charter.

Only targeted interception of traffic and location data in order to combat serious crime – including terrorism – is justified, according to a long-awaited decision by the European court of justice (ECJ) in Luxembourg.

The finding came in response to a legal challenge initially brought by the Brexit secretary, David Davis, when he was a backbench MP, and Tom Watson, Labour’s deputy leader, over the legality of GCHQ’s bulk interception of call records and online messages.

Davis and Watson, who were supported by Liberty, the Law Society, the Open Rights Group and Privacy International, had already won a high court victory on the issue, but the government appealed and the case was referred by appeal judges to the ECJ. The case will now return to the court of appeal to be resolved in terms of UK legislation.


The aim of going to Luxembourg was to clarify EU law on surveillance. The two MPs had argued successfully in the domestic courts that the Data Retention and Investigatory Powers Act (Dripa) 2014 was illegal. The Dripa case was heard by 15 ECJ judges. It coincided with successive atrocities in Paris, Brussels and Nice that reinforced political demands for expansion of powers to intercept emails and phone calls to help catch Islamic State militants operating on the continent.

Lawyers for the UK government maintained that intercepted communications have been at the heart of every terrorist case investigated by police and the security services in recent years.

Responding to the ruling, Watson said: “At a time when we face a real and ever-present terrorist threat, the security forces may require access to personal information none of us would normally hand over. That’s why it’s absolutely vital that proper safeguards are put in place to ensure this power is not abused, as it has been in the recent past.

“Most of us can accept that our privacy may occasionally be compromised in the interests of keeping us safe, but no one would consent to giving the police or the government the power to arbitrarily seize our phone records or emails to use as they see fit. It’s for judges, not ministers, to oversee these powers.”

Martha Spurrier, director of the human rights group Liberty, said: “Today’s judgment upholds the rights of ordinary British people not to have their personal lives spied on without good reason or an independent warrant. The government must now make urgent changes to the Investigatory Powers Act [IPA] to comply with this.

“This is the first serious post-referendum test for our government’s commitment to protecting human rights and the rule of law. The UK may have voted to leave the EU – but we didn’t vote to abandon our rights and freedoms.” Liberty is preparing to challenge the IPA in court.


Jim Killock, executive director of the Open Rights Group, said: “Blanket surveillance of our communications is intrusive and unacceptable in a democracy.


“The government must act quickly to rewrite the IPA or be prepared to go to court again.”

    The full story of MIT biologist Salvador E. Luria’s life spans three countries and two continents. He mentored a lineup of Nobel Prize winners that included James Watson, David Baltimore ‘61, Susumu Tonegawa, and Phillip Sharp, HM ‘96. And his fortunes were altered by ideologies from fascism to McCarthyism to the peace movement. Yet to distill Luria’s role in the scientific triumphs and political turbulence of the 20th century, one need only point to a pair of front-page stories in the New York Times, published within days of each other in October of 1969.

The first story reports Luria’s shared Nobel Prize for pioneering work on bacteriophages (viruses that infect bacteria)―work he began in the United States early in 1941, shortly after fleeing Mussolini’s Italy and Nazi-occupied France. Congratulatory calls and cables poured in, including one from President Richard Nixon. Three days later, Luria learned from another Times story that ?Nixon’s administration had blacklisted him from advising the NIH on scientific grants, apparently because of his vocal role in mobilizing faculty and public sentiment against the Vietnam War.

The blacklisting did not harm Luria’s scientific work, and he soon turned his attention to another war―the so-called war on cancer. In 1972, Luria led the Institute’s effort to secure a $4.4 million grant from the newly formed National Cancer Institute to start the MIT Center for Cancer Research, which he directed from its opening in 1973 until 1985. He championed the idea of understanding the causes of cancer through molecular biology rather than clinical observation, and MIT received funding even though it did not have a medical school. MIT scientists went on to publish research that electrified the field. And Luria, who had become a U.S. citizen in 1947, continued to speak out about political issues of the day.

“I made up my mind that as a citizen, I would be an active participant in American politics, taking advantage of the democratic opportunities that were not available to me in Italy,” he told Time magazine in 1985, six years before his death. “What scientific achievement I have reached is due to the freedom provided in this wealthy country to all aspects of intellectual enterprise.”

Unlike Watson, who was Luria’s first graduate student at Indiana University (and who went on to unlock the structure of DNA at Cambridge with Francis Crick), Luria has never been the subject of a Hollywood biopic. But his life was not short of drama. Born in Turin in 1912 to a lower-middle-class Jewish family, he studied medicine and then decided he wanted to do research. Race laws barred him from a research fellowship in Italy, so he left for Paris in 1938. He studied viruses at the Pasteur Institute until the Nazi invasion in 1940 spurred him to flee to New York aboard the S.S. Nea Hellas. As he says in his autobiography, he had $52 in cash and one suit, making him better off than many of the other 800 refugees on the ship.

In the United States, Luria went on to conduct virus research that shapes the field to this day. His Nobel-winning work, with Max Delbrük and Alfred Hershey, showed that bacteria develop resistance to bacteriophages through random mutations―the same phenomenon that helps drive evolution in more complex organisms. This led to an explosion of research in bacterial genetics. Luria, who’d joined the MIT faculty in 1959 and was named an Institute Professor in 1970, was also the first to discover the restriction-modification phenomenon, in which bacteria protect themselves by altering the DNA of their phage invaders. This work later led to the discovery of so-called restriction enzymes, which would be used for making recombinant DNA and, ultimately, for genetic engineering. He wrote the first comprehensive textbook on modern virology, and he was among the pioneers who hypothesized that some forms of cancer have viral origins. He also sounded the alarm about the “terrifying power” of modern biology, urging that information gleaned from genetic research be used responsibly.

“Luria was both a founder of the new science of molecular biology … and one of the most respected intellects in academic science,” says Sharp, whom Luria recruited to MIT in 1974. (Now an Institute Professor, Sharp succeeded Luria as leader of the cancer center and was named the Salvador E. Luria Professor of Biology.) At MIT, Luria was a protective mentor, shielding his junior faculty members from unnecessary interruptions so they could focus on their research: upon discovering that a construction project would require an associate professor to move out of his office at a critical juncture before his tenure decision, “he went cursing down the hall with the responsible architect in tow,” Sharp recalls. Tonegawa, who won a Nobel in 1987 for his work on the genetic basis of the immune system, joined the cancer center at Luria’s invitation in 1981; Baltimore, who shared a 1975 Nobel for the discovery of reverse transcriptase,Wholesale desktops, first met Luria in 1959, when he was an undergraduate, and calls him the “godfather” of his career.

Sharp says Luria guided him on the internal politics facing faculty members at research institutions―the pursuit of grants, office space, and tenure, as well as more serious challenges. In 1977, he recalls, another scientist had published an article taking credit “in a back-handed fashion” for discovering that genes were sometimes split into several segments instead of making up one continuous segment within a DNA molecule―something that Sharp had discovered earlier that year, and for which he later won the Nobel Prize. “Salva was upset about this and encouraged me to be proactive in countering the claim,” Sharp says. “He had a wonderful sense of politics, either governmental or academic, and enjoyed it.”

Luria’s involvement in national politics, including his endorsement of Henry Wallace’s Communist Party-supported presidential run on the Progressive ticket in 1948, attracted attention from the FBI. A two-year investigation cleared him of any involvement in Communist activity, but nonetheless, he was denied a passport from 1952 to 1959. In the early 1960s, Luria helped found the Boston Area Faculty Group on Public Issues (BAFGOPI), which initially worked to support nuclear disarmament and educate the public on nuclear weapons testing. As U.S. involvement in Vietnam and Cambodia grew, BAFGOPI ran a provocative series of antiwar “ad-itorials” in the New York Times, the Boston Globe, and the Washington Post―including one that stated simply, “Mr. President: STOP THE BOMBING,” and was signed by 2,000 academics from across the United States.

“He was not afraid to call people up and say, ‘I need to raise $10,000 tonight so we can get an ad in the New York Times tomorrow that opposes the bombing of Cambodia,’” Baltimore recalls.

Outside the scientific and political fray, Luria enjoyed poetry and literature. His autobiography is sprinkled with quotations from the likes of T. S. Eliot and William Blake, and for five years he and his wife, Zella, a psychologist, ran an informal literature seminar for MIT biology graduate students in their home, studying works by Dante, Voltaire, and Kafka and the Bhagavad Gita. Luria later hosted a similar seminar with biology professor Frank Solomon. Hong Ma, PhD ‘88, now a professor at Fudan and Penn State universities, reflects that in his native China, “literary discussions focused on understanding the opinions of experts.” When he took part in the workshop with Luria and Solomon, he found it eye-opening that “everyone can comment on the author or the work, without being criticized as being ‘wrong.’”

Luria also took up sculpting at 50, though he acknowledged that a perfectionist streak kept him from pursuing it past his successful first efforts. He claimed to have watched television only once in his life―to see a program featuring a panel of eight American socialists. All the while, he battled depression, a struggle that he wrote candidly about in his 1984 autobiography. “That … was the first time I became aware that he took lithium for depression for most of the later part of his life,” Sharp says. “He was one of the most spontaneous people I knew. When asked about it, he said that depression had almost paralyzed him before this treatment.”

Baltimore says that his mentor was truly a humanist: “That’s what drove him, really, a sense of what’s right and wrong and human need.” And from the final words of his autobiography, it seems that Luria would concur: “The story of one’s life path acquires humane significance from the contacts with other human beings. Dante’s allegory, seeking insight and understanding in the words of the dwellers of Hell, Purgatory, and Heaven, is also an allegory of life deeply shared. Even as one approaches the end of the journey, one hopes, like Dante, for one final stretch of meaningful toil.”

混凝槽

螺旋轴
  中心轴为高温高压整体锻造制成,保证轴的结实耐用,YCB不锈钢圆弧泵厂家。螺旋叶片表面做耐磨堆焊和热处理工艺,接触面维氏硬度可达到800度以上。螺旋轴使用大型车床车削加工,保证轴的同心和精度;二次特殊车床修磨校正堆焊层,还起到光滑堆焊表面的作用,减少磨擦。 
    
垫片
  都是数控车床加工而成,固定环与活动环之间公差在0.02mm以内。 
    
动定环片
  采用德国通快激光切割机切割而成,精度高,且经过抛光处理增长使用寿命。按客户特殊使用要求,可以进行热处理,使环片维氏硬度增加1300至1500度(常规材料表只有990度),更加加强了环片耐腐蚀性能,使得环片不与氯离子反应而被腐蚀。

  可通过变频器调节,转速为1到4转/min,减速装置选用低噪音、高效率的国内外知名品牌产品,保障质量稳定和维修方便。
